Purity assessment relies mainly on reversed-phase high-performance liquid chromatography, which separates the target heptapeptide from truncated sequences, deletion analogues, and oxidised forms. Mass spectrometry, usually coupled to liquid chromatography, confirms identity through the expected molecular ion and reveals modifications such as methionine sulfoxide formation. Amino acid analysis can verify composition, and tandem mass spectrometry supports sequence confirmation. Ultraviolet detection near 254 to 280 nanometres is convenient because the phenylalanine and histidine residues absorb in that region. Nuclear magnetic resonance is rarely used for routine release testing.
Common degradation pathways include oxidation of the methionine side chain, hydrolysis of the peptide backbone, and aggregation under unfavourable pH or concentration. Stability studies typically monitor the main peak by chromatography and report total related substances as a percentage. Because no official monograph exists, acceptance criteria vary between laboratories, and reported purity values are not directly comparable across suppliers. Analysts therefore document the method, column, and detection wavelength alongside each result, and open questions remain about how much biological activity the oxidised forms retain.
Solid semax is typically supplied as a lyophilised powder that is hygroscopic and sensitive to moisture, light, and repeated temperature cycling. Long-term storage of the dry peptide is generally recommended at approximately -20 degrees Celsius, while shorter working periods may use refrigeration at 2 to 8 degrees Celsius. Vials should remain tightly closed and desiccated when brought to room temperature, because condensation can damage the material before it is weighed. Dividing a batch into aliquots is preferable to thawing one container repeatedly.
The peptide is prone to several degradation pathways. Oxidation of the methionine residue produces a sulfoxide that elutes close to the parent peak in many chromatographic systems. Hydrolysis of peptide bonds and deamidation of susceptible residues in related sequences also reduce purity over time. Lyophilised material kept dry at minus twenty degrees Celsius and shielded from light is the most stable form commonly described in laboratory practice.
Material sold for laboratory use varies widely in stated purity and documentation. A certificate of analysis should list the analytical method, the column and detector used, and the observed purity value. Independent verification by an outside laboratory is the practical way to confirm identity when documentation is absent or internally inconsistent. Regulatory status differs by country, and a product legal in one jurisdiction may be unapproved or controlled in another.
Identity and purity of semax are established with reversed-phase high-performance liquid chromatography coupled to ultraviolet detection, usually at 214 nanometres. Mass spectrometry, most often electrospray ionisation in positive mode, confirms the molecular mass and reveals truncated sequences. Amino acid analysis and peptide mapping after enzymatic digestion provide additional structural confirmation. Laboratories typically report purity as the percentage area of the main peak, a figure that does not capture isomeric or oxidised variants unless the method resolves them.
| Property | Value | Notes |
|---|---|---|
| Typical analytical method | Reversed-phase HPLC with UV detection | Separates target peptide from truncated and oxidised forms |
| Identity confirmation | LC-MS or ESI-MS | Compares observed and calculated molecular ion |
| Primary degradation route | Methionine oxidation | Forms the sulfoxide; monitored as an early-eluting peak |
| Short-term storage | 2 to 8 degrees Celsius | Applies to working vials and reconstituted portions |
| Common synonyms | ACTH(4–10) analogue, MEHFPGP | Descriptive names used in laboratory catalogues |
Lyophilized material is chemically stable for extended periods when kept dry, cold, and protected from light. The powder is hygroscopic, so vials should be warmed to room temperature before opening to reduce condensation on the contents. Once dissolved, the peptide is far less stable because peptide bonds are susceptible to hydrolysis and the methionine residue can oxidize. Solutions are typically aliquoted and held at 2-8 °C for short intervals or frozen for longer ones, and repeated freeze-thaw cycles should be avoided.
Routine characterization relies on reversed-phase high-performance liquid chromatography to establish purity and on mass spectrometry to confirm molecular identity. Electrospray ionization and matrix-assisted laser desorption ionization are both used for mass verification. Amino acid analysis and peptide mapping can detect sequence errors. Common impurities include truncated sequences, methionine sulfoxide formed by oxidation, and deamidated products. Chromatograms are usually recorded near 214 nm, where the peptide backbone absorbs, and purity is reported as the percentage area of the principal peak.
Scarborough is represented by six ridings for the provincial government and Federal government. Municipal riding boundaries were harmonized within the City of Toronto to match the provincial boundaries in 1999 through provincial legislation called The Fewer Municipal Politicians Act of 1999. This took effect on December 1, 2000. Ridings were represented by two councillors per riding until 2018 when the Government of Ontario reduced this to one councillor per riding. Scarborough now has six councillors.
== Function == The basal lamina is made and maintained by the cells that sit on it. It acts as a point of attachment for cells. However, it can also have other function such as a permeability barrier in the glomerulus (urine production). Some of the matrix molecules (of the basal lamina) mediate synaptic adhesion in neuromuscular synapses. Alport syndrome is a genetic disorder resulting from mutations in the COL4A3/4/5 genes. These genes are important in collagen IV synthesis and basement membrane formation. In individuals with this syndrome the basement membrane in structures such as the glomerulus, ears, and eyes does not function properly, causing symptoms such as blood in the urine, loss of hearing, and vision problems.
== Life and work == Magati was born in Scandiano, Reggio Emilia, in the landed family of Giorgio and Laura Mattacoda. A brother, Giovanni Battista, became a physician while a sister became the grandmother of Antonio Vallisneri. He studied at Pauda and from 1596, medicine at the University of Bologna. Graduating in 1597 he worked at the Hospital of Santa Maria della Consolazione in Rome. He was influenced by the teachings of Flaminio Rota, Giulio Cesare Claudini, and Giovanni Battista Cortese. He then took the exam of the College of Physicians and became a surgeon at the Hospital of Santa Anna. He then returned to Scandiano and around 1612, through the influence of Marquis Enzio Bentivoglio, he became a lecturer in surgery at Ferrara. In 1618 he became very ill and gave up teaching. He joined the Capuchin order as a lay brother in 1618 and took his vows in Ravenna the next year, and going by the name of Padre Liberato da Scandiano. He continued to practice medicine for the House of Este. In 1647 Magati was operated on for gall-stones at Bologna but he died three days after the surgery. Magati's major contribution was in wound hygiene and healing. He went against the contemporary practice of frequent change in dressing and the use of ointments. He instead suggested that natural processed played a key role in healing and that these processes needed to be aided. For this he is remembered as a fundamental reformer of surgery.
Although many natural toxins are secondary metabolites, these poisons also include peptides and proteins. An example of a toxic peptide is alpha-amanitin, which is found in relatives of the death cap mushroom. This is a potent enzyme inhibitor, in this case preventing the RNA polymerase II enzyme from transcribing DNA. The algal toxin microcystin is also a peptide and is an inhibitor of protein phosphatases. This toxin can contaminate water supplies after algal blooms and is a known carcinogen that can also cause acute liver haemorrhage and death at higher doses. Proteins can also be natural poisons or antinutrients, such as the trypsin inhibitors (discussed in the "metabolic regulation" section above) that are found in some legumes. A less common class of toxins are toxic enzymes: these act as irreversible inhibitors of their target enzymes and work by chemically modifying their substrate enzymes. An example is ricin, an extremely potent protein toxin found in castor oil beans. This enzyme is a glycosidase that inactivates ribosomes. Since ricin is a catalytic irreversible inhibitor, this allows just a single molecule of ricin to kill a cell.

The first two bus operating contracts for the new Weaver Network were awarded in September 2026 to FirstGroup, covering Huddersfield and parts of Leeds, and Transdev Blazefield, covering Dewsbury. In October 2021, £830 million of funding was announced for the West Yorkshire Combined Authority through the City Region Sustainable Transport Settlement, including funding to develop mass transit proposals for the region. Consultation was undertaken between July and September 2024 on proposed tram routes linking Bradford and Leeds, and St. James Hospital, central Leeds and White Rose Shopping Centre. Following a review by the National Infrastructure and Service Transformation Authority, the programme was moved to a more sequential approach intended to reduce delivery risk, contributing to a delay in the expected timetable for the scheme.
Protein DAMPs include intracellular proteins, such as heat-shock proteins or HMGB1, and materials derived from the extracellular matrix that are generated following tissue injury, such as hyaluronan fragments. Non-protein DAMPs include ATP, uric acid, heparin sulfate and DNA.
=== Surface of the Moon === In 2009, India's Chandrayaan-1 satellite and the National Aeronautics and Space Administration (NASA) Cassini spacecraft and Deep Impact probe each detected evidence of water by evidence of hydroxyl fragments on the Moon. As reported by Richard Kerr, "A spectrometer [the Moon Mineralogy Mapper, also known as "M3"] detected an infrared absorption at a wavelength of 3.0 micrometers that only water or hydroxyl—a hydrogen and an oxygen bound together—could have created." NASA also reported in 2009 that the LCROSS probe revealed an ultraviolet emission spectrum consistent with hydroxyl presence. On 26 October 2020, NASA reported definitive evidence of water on the sunlit surface of the Moon, in the vicinity of the Clavius crater, obtained by the Stratospheric Observatory for Infrared Astronomy (SOFIA). The SOFIA Faint Object infrared Camera for the SOFIA Telescope (FORCAST) detected emission bands at a wavelength of 6.1 micrometers that are present in water but not in hydroxyl. The abundance of water on the Moon's surface was inferred to be equivalent to the contents of a 12-ounce bottle of water per cubic meter of lunar soil. The Chang'e 5 probe, which landed on the Moon on 1 December 2020, carried a mineralogical spectrometer that could measure infrared reflectance spectra of lunar rock and regolith. The reflectance spectrum of a rock sample at a wavelength of 2.85 micrometers indicated localized water/hydroxyl concentrations as high as 180 parts per million.

When fellow guard Bosch dissents, Barris pressures him to continue. Travis discovers that Benjy, now severely ill, concealed his need for insulin, believing he could cure his diabetes merely through dieting. Bosche tries to help find Benjy's insulin, but is caught by other guards. Barris provides Benjy's insulin, but later has all the guards beat Bosche severely and orders Travis to clean the prison toilets. When Travis taunts Barris, the guards respond by shoving his head into the toilet, nearly drowning him. One morning during roll call, Travis removes his shirt as a sign of protest, followed by the other prisoners. He climbs up to one of the cameras and demands they be released, but the guards choke him. When Benjy tries to defend Travis, Barris bludgeons him. Guards lock Travis into an old boiler pipe overnight, attack the remaining prisoners, and handcuff each man across the cell doors. While locked in the boiler, Travis discovers a hidden infrared camera. As his despondency turns to anger, he manages to escape and interrupts a guard’s attempt to rape a prisoner. The intended victim and Travis beat the guard and knock him out before freeing the other prisoners. Finding Benjy dead from his head injury, Travis leads an assault against the guards, chasing them through the building. As the remaining guards try to lift the garage door to escape, Barris tries to keep them in, unwilling to forfeit his power. A vicious brawl ensues with the prisoners overwhelming the guards.
=== 1987 === 19 June: Heather West (16). Heather was likely to have been murdered because the Wests considered her efforts to leave the household as a threat, since she had divulged to her classmates the extensive physical and sexual abuse that occurred at Cromwell Street. Fred claimed he had not intended to kill his daughter, but carpet fibres found on two lengths of rope discovered with her remains suggested that she had been restrained and subjected to a sexual assault prior to her murder. Her body was dismembered with a heavy serrated knife and later buried in a hole in the garden, which Fred got his son to dig under the pretence of installing a fish pond. The 1994 police investigation into Heather's disappearance led to the discovery of her body and the arrest of both her parents.

Dry powder is usually kept at about -20 degrees Celsius for long-term storage and at 2 to 8 degrees Celsius for short working periods. Containers should stay sealed and protected from light and moisture to limit degradation.
Oxidation of the methionine residue to the sulfoxide is a frequently reported change, and it usually appears as an earlier-eluting peak in reversed-phase chromatography. Backbone hydrolysis products can also accumulate in aqueous solution.
Liquid chromatography coupled with mass spectrometry is the usual approach, because the observed molecular ion can be compared with a calculated mass of about 813.9 Da. Retention time alone does not establish identity.
Reversed-phase high-performance liquid chromatography is the standard method, with detection in the ultraviolet range. Peak area percentage yields a purity figure for the main component. Mass spectrometry is normally run alongside to confirm molecular identity.
