However it was later withdrawn from the market in February 2020 due to a higher risk of malignancy in a randomized trial of lorcaserin. Previously approved agents were subsequently removed from the US market. Lorcaserin is a full agonist for 5-HT2C and 5-HT2B receptors and partial agonist for 5-HT2A receptors (75% of the maximal response elicited by serotonin). Lorcaserin is a potent and selective 5-HT2C agonist with rapid oral absorption that shows dose-dependent decrease in food intake and body weight. Lorcaserin affects body weight by producing a negative energy balance through reduced food intake (energy intake) without alterations in energy expenditure and substrate oxidation. Lorcaserin has a high affinity for the 5-HT2C receptors, with 18-fold selectivity over 5-HT2A receptors and 104-fold over 5-HT2B receptors. === Upgrades === Chain Home was the primary radar system of the UK for only a short time. By 1942, many of its duties had been taken over by the far more advanced AMES Type 7 ground-controlled interception (GCI) radar systems. Whereas CH scanned an area perhaps 100 degrees wide and required considerable effort to take measurements, the Type 7 scanned the entire 360-degree area around the station, and presented it on a plan position indicator, essentially a real-time two-dimensional map of the airspace around the station. Both fighters and bombers appeared on the display, and could be distinguished using Identification friend or foe (IFF) signals. The data from this display could be read directly to the intercepting pilots, without the need for additional operators or control centres. With the deployment of GCI, CH became the early warning portion of the radar network. To further simplify operations and reduce manpower requirements, the job of plotting the targets became semi-automated. An analogue computer of some complexity, known as "The Fruit Machine", was fed information directly from the operator console, reading the goniometer setting for bearing, and the range from the setting of a dial that moved a mechanical pointer along the screen until it lay over a selected target. When a button was pushed, the Fruit Machine read the inputs and calculated the X and Y location of the target, which a single operator could then plot on a map, or relay directly over the telephone.
=== English === Order, Sons of St. George - Organized after the Civil War by Englishmen in the anthracite coal-mining region of Pennsylvania to counter the Molly Maguires. The organization "took permanent shape" at Scranton in 1871. Membership was open to "Englishmen, their sons, and grandsons, wherever born". Beneficiary membership was limited to those between 18 and 50; those over 50 were allowed honorary membership. The Order required belief in a Supreme Being, reverence for the Holy Bible, and loyalty to one's adopted country. By 1896 the order had about 35,000 members in the United States, Canada, and Hawaii. Ritual based on the legend of St. George included a "language of words, signs and grips" that the member learned upon initiation which could identify him to other members of the order. The Orders emblem was St. George conquering the dragon. The system of sick benefits varied by the lodge and the inclination of members. There was also a funeral benefit for members and their wives and a benevolent fund for brethren and "any worthy Englishman in distress". Some lodges also provided physicians and medicine for sick members. There was also a female auxiliary, the Daughters of St. George, but it was not officially recognized by the Supreme Lodge. In 1923 the Order was accused of promoting pro-British propaganda in textbooks used in New York by a representative of Mayor John Francis Hylan. === Screening of DELs in cells === DNA-encoded libraries (DELs) have been adapted for screening in living cells to better reflect native biological conditions, specifically using Xenopus laevis oocytes. This approach, termed cellular Binder Trap Enrichment (cBTE), facilitates the identification of small-molecule ligands that bind to target proteins in a native cellular environment. In this method, the protein of interest (POI) is expressed in oocytes as a fusion with a "Prey" protein, such as carbonic anhydrase IX (CAIX). Simultaneously, a "Bait" molecule—comprising a known ligand for the Prey protein linked to a DNA strand—is introduced. Alongside the Bait, a DEL is co-injected into the oocytes. If a DEL member binds to the POI, it brings its attached DNA tag into the same molecular complex as the Bait DNA via the POI–Prey–Bait interaction. Following incubation, the oocytes are lysed, and the lysate is subjected to Binder Trap Enrichment (BTE) as described above. In essence, the DEL and Bait DNA are ligated in droplets, thus encoding the binding event. The ligated DNA is then amplified and subjected to high-throughput sequencing to identify the DEL members that interacted with the POI.
