== In cancer progression and metastasis == Initiation of metastasis requires invasion, which is enabled by EMT. Carcinoma cells in a primary tumor lose cell-cell adhesion mediated by E-cadherin repression and break through the basement membrane with increased invasive properties, and enter the bloodstream through intravasation. Later, when these circulating tumor cells (CTCs) exit the bloodstream to form micro-metastases, they undergo MET for clonal outgrowth at these metastatic sites. Thus, EMT and MET form the initiation and completion of the invasion-metastasis cascade. At this new metastatic site, the tumor may undergo other processes to optimize growth. For example, EMT has been associated with PD-L1 expression, particularly in lung cancer. Increased levels of PD-L1 suppresses the immune system which allows the cancer to spread more easily. === Purpose === There are two major applications to studying the genome at the single-cell level. One application is to track the changes that occur in bacterial populations, where phenotypic differences are often seen. These differences are easily missed by bulk sequencing of a population, but can be observed in single-cell sequencing. The second major application is to study the genetic evolution of cancer. Since cancer cells are constantly mutating it is of great interest to researchers to see how cancers evolve at the level of individual cells. These patterns of somatic mutations and copy number aberration can be observed using single-cell sequencing.
=== Peltier === Peltier refrigerators are powered by electricity, usually 12 volt DC, but mains-powered wine coolers are available. Peltier refrigerators are inexpensive but inefficient and become progressively more inefficient with increased cooling effect; much of this inefficiency may be related to the temperature differential across the short distance between the "hot" and "cold" sides of the Peltier cell. Peltier refrigerators generally use heat sinks and fans to lower this differential; the only noise produced comes from the fan. Reversing the polarity of the voltage applied to the Peltier cells results in a heating rather than cooling effect. Other specialized cooling mechanisms may be used for cooling, but have not been applied to domestic or commercial refrigerators.

In September 2008, a Green Beret ODA conducted a joint operation with 1 Troop 3 Squadron SASR in northwest Uruzgan Province, the operation was designed to lure insurgents into a trap using a ground convoy of five special forces GMV trucks as bait. SASR sniper teams, who inserted on foot the night before as part of two cut-off groups to overwatch the patrol. The plan worked, a small group of insurgents approached the vehicles, intent on ambushing the convoy, they were engaged by the SASR snipers. Minutes later a Toyota Hilux appeared carrying a number of armed insurgents, they were engaged and killed and a second vehicle, a van, arrived carrying 3 insurgents were engaged by the sniper teams until they spotted a female non-combatant who was used as a human shield by the surviving insurgent, the insurgent was eventually killed with no harm to the woman, a total of 13 insurgents were killed.

=== Improvements in forensics === With the advent of and improvements to DNA testing/DNA profiling and other forensics technology, many cold cases are being re-opened and prosecuted. Police departments are opening cold case units whose job is to re-examine cold case files. DNA evidence helps in such cases but as in the case of fingerprints, it is of no value unless there is evidence on file to compare it to. However, to combat that issue, the FBI is switching from using the Integrated Automated Fingerprint Identification System (IAFIS) to using a newer technology called the Next Generation Identification (NGI). == As an oncolytic agent == Sendai virus-based anticancer therapy for model and companion animals has been reported in several scientific papers. The described studies demonstrate that Sendai virus has a potential of becoming a safe and effective therapeutic agent against a wide range of human cancers. High genomic stability of SeV is a very desirable trait for oncolytic viruses. SeV is not likely to evolve into a pathogenic strain or into a virus with decreased oncolytic potential. The cytoplasmic replication of the virus results in a lack of host genome integration and recombination, which makes SeV safer and more attractive candidate for broadly used therapeutic oncolysis compared to some DNA viruses or retroviruses.
