=== Fraction collector === The fraction collector is typically a rotating rack that can be filled with test tubes or similar containers. In large FPLC columns the sample may be loaded into the column directly using a small peristaltic pump rather than an injection loop. When the buffer contains dissolved gas, bubbles may form as pressure drops where the buffer exits the column; these bubbles create artifacts if they pass through the flow cells. This may be prevented by degassing the buffers, e.g. with a degasser, or by adding a flow restrictor downstream of the flow cells to maintain a pressure of 1-5 bar in the eluant line.

== Mechanism of action == Endorphins are released from the pituitary gland, typically in response to pain, and can act in both the central nervous system (CNS) and the peripheral nervous system (PNS). In the PNS, β-endorphin is the primary endorphin released from the pituitary gland. Endorphins inhibit transmission of pain signals by binding μ-receptors of peripheral nerves, which block their release of neurotransmitter substance P. The mechanism in the CNS is similar but works by blocking a different neurotransmitter: gamma-aminobutyric acid (GABA). === Cap-dependent translation initiation === The eukaryotic translation initiation factor eIF4E plays a central role in directing ribosomes to the 5′-cap structure of mRNAs, thereby facilitating efficient protein synthesis. Vegan diets are often higher in iron than lacto-ovo vegetarian diets, because dairy products contain very little iron and can displace iron-containing foods from the diet. The first FDA-approved therapeutic monoclonal antibody was a murine IgG2a CD3 specific transplant rejection drug, OKT3 (also called muromonab), in 1986. This drug found use in solid organ transplant recipients who became steroid resistant. Hundreds of therapies are undergoing clinical trials. As of 2025, there were over 200 antibodies approved by the FDA, with about 40% of them targeting oncological targets and 35% for autoimmune and inflammatory diseases. The majority of these are conventional antibodies which make up over 70% of FDA approved antibodies. Most are part of the IgG1 subclass, again making up over 70% of antibodies.
=== Isolation === Many biologically active natural compounds are derived from fungi, and guanacastepene A is one such compound. Attempts were made to characterize CR115, but CR115 did not form spores under any of the conditions tested, and no other morphological characteristics were observed that provided insight into its phylogeny. rDNA sequence analysis shows a similarity of 90% to an uncharacterized root basidiomycete. The fungal strain was cultivated in potato dextrose broth for 14–21 days. The culture was then extracted with hexane to obtain a crude extract containing nonpolar secondary metabolites. This extract was fractionated by C18 flash column chromatography using an acetonitrile/water (CH3CN/H2O) step gradient and subsequently purified by reverse-phase HPLC.

Light-emitting diode therapy (LEDT) is a clinical approach that applies different wavelengths of light to cure diseases or conditions with skin-safe lights. Following NASA's innovation in the 1990s with Light Emitting Diodes (LEDs) that emit a specific narrow light spectrum, LED Therapy (LEDT) showed significant potential. The high precision of narrow-band LED therapy enabled its first use in clinical practices. The commonly used lights in LEDT are blue, red, green, yellow, and infrared (IR). LEDT's general mechanism is related to cellular receptor metabolism. Light functions as an external stimulus and influences cellular metabolism by initiating photo-biochemical reactions within cells. Light Emitting Diode Therapy (LEDT) encompasses two primary therapeutic approaches: photodynamic Therapy (PDT) and photobiomodulation Therapy (PBMT). Photodynamic therapy (PDT) utilises light-sensitive compounds combined with LED light to generate reactive oxygen species, which selectively target and destroy abnormal cells. Oncology and certain skin conditions widely use this technique. Whereas photobiomodulation therapy (PBMT) utilizes low-level LED light to stimulate cellular repair, stimulate wound healing, and reduce inflammation, without the use of photosensitizing agents. Different wavelengths and mechanisms are utilized for different therapeutic effects.
